What it is
SNAP-8 is acetyl octapeptide-3, a topical cosmetic peptide that interferes with the SNARE complex to reduce acetylcholine release at the neuromuscular junction. It is the eight-residue peer of our existing argireline record. The honest note is the value here: its efficacy figures come from manufacturer-funded studies, and real-world results are considerably smaller.
How it works
Muscle contraction requires acetylcholine to be released from vesicles at the neuromuscular junction, and that release depends on the SNARE complex assembling correctly. SNAP-8 competitively mimics the N-terminal end of SNAP-25, one of the SNARE proteins, interfering with complex formation and reducing the efficiency of vesicle exocytosis. Less acetylcholine released means weaker facial muscle contraction and, in principle, softer expression lines. The unanswered question is delivery: an eight-amino-acid peptide has to cross the stratum corneum to reach anything, and that is precisely what the independent permeability literature exists to test.

The open question
Whether an eight-amino-acid peptide crosses the stratum corneum in a meaningful quantity at all. There is an independent review that addresses exactly this: Zdrada-Nowak et al., International Journal of Molecular Sciences 2025, PMID 40565185, on skin permeability and efficacy of acetyl hexapeptide-8. We could not retrieve it. It is the highest-value outstanding follow-up for this record and for our argireline record both.
One caution: some vendors sell an injectable "SNAP-8". There is no evidence for that route at all.
