Why this tier

Khavinson demonstrated telomerase activation and telomere elongation in human somatic cells in vitro, and Anisimov extended rodent lifespan by 10–25% with reduced spontaneous tumour incidence, but there is no human longevity or clinical outcome trial. FDA reports zero FAERS adverse-event reports for it through December 2025, which FDA itself footnotes as a reflection of how little anyone reports, not as evidence of safety.

Overview

What it is

Epitalon is a synthetic four-amino-acid peptide (alanyl-glutamyl-asparagyl-glycine) developed by Vladimir Khavinson at the Saint Petersburg Institute of Bioregulation and Gerontology. It is based on epithalamin, a natural pineal gland peptide, and targets telomere shortening. It is run in short periodic courses of 10–20 days rather than continuously.

Mechanism

How it works

Every time a cell divides, the protective telomere caps on the ends of its chromosomes get slightly shorter. When they become critically short the cell stops dividing and enters senescence, which contributes to tissue ageing. Telomerase is the enzyme that can rebuild telomere length, and its activity declines with age. Khavinson's group demonstrated that Epitalon activates telomerase in human somatic cells, producing telomere elongation. In a fibroblast study, treated cells showed increased telomerase activity and divided beyond the normal Hayflick limit. The second mechanism is pineal: Epitalon stimulates the pineal gland to maintain healthy melatonin production, which declines with age and affects sleep quality, circadian rhythm and antioxidant defence.

    Reported as a short course, 2–3 times per year, spaced 4–6 months apart, working out to roughly 100 mg across a cycle. The two shapes reported are 5 mg for 20 days or 10 mg for 10 days. Effects on telomerase activity are described as persisting beyond the dosing window. One caution about where the human end of this comes from: the tier-1 courses were given as epithalamin, the bovine pineal extract, not as the synthetic tetrapeptide, and they come from a single unreplicated Russian group. One outlying source gives 2 mg/day, five times lower.

How it got here

  1. 22 April 2026

    FDA removed 12 peptide bulk substances from Category 2 after the nominators withdrew their nominations. Announced 15 April 2026, effective 22 April 2026. Removal from Category 2 is not permission to compound.

  2. 24 July 2026

    PCAC voted in favour of recommending Epitalon for the 503A bulks list. Seven members voted in favour with one abstention; sources disagree on whether four or five voted against, and FDA has not published the minutes, so we publish no tally.

An advisory vote is not law. In July 2026 an FDA advisory committee voted in favour of adding several peptides to the 503A bulks list, over the explicit recommendation of FDA’s own scientific reviewers, who recommended against all seven. FDA would still have to write a rule, and it has not proposed one or given a date. None of these compounds is lawfully compoundable today.Note what FDA actually assessed: BPC-157 for ulcerative colitis, TB-500 for wound healing, KPV for wound healing and inflammatory conditions, MOTS-c for obesity and osteoporosis, Epitalon for insomnia, Semax for cerebral ischaemia, migraine and trigeminal neuralgia. Whatever emerges from rulemaking will be scoped to those indications.

The 2026 Prohibited List entered force on 1 January 2026.
We do not state a status

Not named on WADA’s 2026 Prohibited List. Its S0 section also bans drugs it does not name, when no government health authority currently approves them for human treatment. We found no such approval for epitalon. USADA, the US anti-doping agency, warns that many experimental peptides are prohibited in sport. So athletes should treat epitalon as likely prohibited and check with their federation before competing.

The FDA reclassification has no effect whatsoever on WADA status.

What it is

A synthetic tetrapeptide of four amino acids, alanyl-glutamyl-asparagyl-glycine, developed by Vladimir Khavinson at the Saint Petersburg Institute of Bioregulation and Gerontology. It is derived from epithalamin, a natural peptide extracted from the pineal gland.

Two mechanisms

The telomere mechanism is the headline. Each cell division shortens the telomere caps protecting the chromosome ends. Critically short telomeres push the cell into senescence, and that accumulation is one of the recognised hallmarks of ageing. Telomerase rebuilds telomere length but its activity declines with age. Khavinson's group showed Epitalon activates telomerase in human somatic cells, with treated fibroblasts dividing beyond the normal Hayflick limit.

The pineal mechanism is why Epitalon shows up in our sleep guide as well as our anti-aging guide. It stimulates the pineal gland to maintain melatonin production, which declines with age and drives circadian disruption. That makes it the sleep peptide of choice for timing problems: shift work, jet lag, age-related circadian decline. DSIP is the choice for sleep depth.

What the evidence actually supports

In vitro telomerase activation in human cells. In rodents, Anisimov's studies extended lifespan by 10–25%, reduced spontaneous tumour incidence, and improved ageing biomarkers across organ systems.

There is no human longevity study, and no clinical outcome trial. Our anti-aging guide characterises the evidence as moderate: preclinical plus animal longevity data.

How it is run

5–10 mg subcutaneously daily, for 10–20 consecutive days, two to three times per year, spaced four to six months apart. It is a course, not a continuous protocol. The effects on telomerase are described as persisting beyond the dosing window.

That short-course structure makes it relatively cost-effective despite a high per-milligram price, and it is why our anti-aging guide places it as the periodic layer in a longevity stack alongside continuous topical GHK-Cu and cycled MOTS-C.

What we do not know

Our sources publish no side effect profile, no contraindication list beyond general research-peptide caution, and no reconstitution table for Epitalon. Absence from those lists is not evidence of safety. It is absence of research.
