Why this tier

The kisspeptin evidence base is real, intravenous, and mostly about a different molecule. The widely cited desire-and-arousal trials from Imperial College used kisspeptin-54 at 1 nmol/kg/h infused over 75 minutes: in women with HSDD (JAMA Network Open, 26 October 2022, 32 premenopausal women completing both visits) modified fMRI activation to erotic stimuli, LH +2.14 IU/L, no significant change in downstream sex steroids, and a modest exploratory increase in self-reported sexiness. The men’s study from the same group reported penile tumescence 56% above placebo; the peptide form was not independently confirmed and we assume KP-54 by group convention while marking it unverified. Kisspeptin-10 has its own human pharmacology, all intravenous, and there is no human trial evidence for subcutaneous kisspeptin-10 at all.

Overview

What it is

Kisspeptin-10 is a ten-amino-acid fragment that stimulates gonadotropin-releasing hormone neurons and, downstream, luteinising hormone and testosterone. Every human dataset for it is intravenous, and its plasma half-life is four minutes. The widely cited desire-and-arousal trials used a different molecule: kisspeptin-54, which has a half-life of 27.6 minutes. The market sells KP-10 while citing them.

Mechanism

How it works

Kisspeptin sits at the top of the reproductive hormone axis. It acts on kisspeptin receptors on gonadotropin-releasing hormone neurons in the hypothalamus, which raises GnRH pulse output; GnRH then drives luteinising hormone and follicle-stimulating hormone from the pituitary, and LH drives testosterone from the testes. That cascade is well documented in humans by intravenous infusion. Two details matter more than the cascade itself. First, the dose-response is non-monotonic: in men, an intravenous bolus of 1 mcg/kg produced maximal LH stimulation and 3 mcg/kg produced a smaller response than 1 mcg/kg. More is not more. Second, kisspeptin-10 has a plasma half-life of four minutes, which constrains what any single subcutaneous injection can plausibly do.

The correction that justifies this record

The famous kisspeptin trials, the ones every vendor page links to showing changes in brain response to erotic stimuli and improvements in self-reported desire, used kisspeptin-54.

The product being sold is kisspeptin-10.

They are different molecules with different pharmacokinetics. Kisspeptin-10 has a plasma half-life of 4 minutes. Kisspeptin-54 has a half-life of 27.6 minutes, nearly seven times longer. In the trials, KP-54 was not given as a single injection either; it was infused continuously for 75 minutes.

What the desire trials actually found

Women with HSDD, JAMA Network Open, 26 October 2022: kisspeptin-54 at 1 nmol/kg/h infused intravenously over 75 minutes. 40 randomised, 32 premenopausal women completing both visits, mean age 29.2. The findings: modified fMRI activation to erotic stimuli, LH up 2.14 IU/L, FSH up 0.28 IU/L, no significant change in downstream sex steroids, and a modest exploratory increase in self-reported "sexiness". Kisspeptin was well tolerated with no reported adverse effects.

Read that carefully. The primary outcomes are brain imaging and pituitary hormones. The subjective effect is exploratory and modest. Testosterone and oestrogen did not move.

Men with HSDD, same Imperial College group: 32 men, mean age 37.9, double-blind two-way crossover, 75-minute infusion, penile tumescence 56% above placebo, no adverse events. The peptide form was not independently confirmed in our checking. We assume KP-54 by the group's convention and mark it unverified.

Kisspeptin-10's own pharmacology

It exists, and it is worth knowing, because it contains a lesson.

JCEM 2011 (PMID 21632807), healthy men: an intravenous bolus of 1 mcg/kg produced maximal LH stimulation, taking LH from 4.1 ± 0.4 to 12.4 ± 1.7 IU/L at 30 minutes. A 3 mcg/kg bolus produced a smaller response than 1 mcg/kg. The dose-response curve turns over. Tripling the dose made it work less well.

Infusion at 4 mcg/kg/h for 22.5 hours: LH from 5.4 to 20.8 IU/L, testosterone from 16.6 ± 2.4 to 24.0 ± 2.5 nmol/L. At 1.5 mcg/kg/h: LH from 5.2 to 14.1 IU/L, with pulse frequency rising from 0.7 to 1.0 per hour.

So kisspeptin-10 genuinely drives the reproductive axis in humans, by continuous intravenous infusion, in a research setting, over hours.

What that means for how it is sold

There is no human trial evidence for subcutaneous kisspeptin-10. That is the route the community uses.

And a four-minute plasma half-life makes the standard framing, 100 mcg subcutaneously before a workout for a sustained effect, pharmacologically implausible on its face. Whatever a subcutaneous injection of a four-minute peptide does, it is not what a 22-hour infusion did.

Kisspeptin-10 is still in Category 2 under 503A, added 29 September 2023, on immunogenicity grounds and for absent or limited safety information. It is one of the few compounds in this library carrying a live FDA "significant safety risk" designation, and it was not part of the April 2026 removals.
